Autism and the MMR vaccine

Science Tuesday: Back into the hornets nest is a thoughtful follow-up post on the decision of a scientist to vaccinate his child.

Autism isn’t like tuberculosis, there’s not a bacteria that causes the disease. In fact,most researchers believe that “autism” is not a discrete disorder, rather “autism is a clinically defined pervasive developmental disorder with phenotypically diverse neuropsychiatric symptoms and characteristics. These manifest as a spectrum of social and communicative deficits, stereotypical patterns and disturbances of behaviour.”¹

If a particular trait’s heritability is 100% then the trait is due entirely to genetic variation, if the heritability is 0% then the trait is due entirely to environmental variation. By some estimates, heritability of autism spectrum disorders exceeds 90%

repeated studies have found that autism diagnoses continue to rise even after the removal of thimerosal from the vaccine.

Finally, when thinking about the environmental influences on autism, it’s important to explore the role of the environment on genetics. Many of the types of genetic changes that have been identified as causative in autism are indicative of some sort of DNA damage – DNA damage that may result from exposure to an environmental toxin. Many scientists, and I count myself in their number, feel that the recent autism ‘epidemic’ is due primarily to improved screening and diagnosis. In other words, prior to the 1980’s, many people suffering from autism were diagnosed as “slow” or misdiagnosed with another type of mental retardation. Unfortunately, there is no way to quantify this hypothesis.

Using Spice-based Compound To Kill Cancer Cells

Synthetic molecules, derived from curcumin, a naturally occurring compound found in the spice turmeric have been killed cancer cells, in lab settings. Centuries of anecdotal evidence and recent scientific research suggest curcumin has multiple disease-fighting features, including anti-tumor properties. However, when eaten, curcumin is not absorbed well by the body. Instead, most ingested curcumin in food or supplement form remains in the gastrointestinal system and is eliminated before it is able to enter the bloodstream or tissues.

James Fuchs, assistant professor of medicinal chemistry and pharmacognosy at Ohio State University and principal investigator on the project, and colleagues are continuing to refine compounds that are best structured to interact with a few overactive proteins that are associated with cell activity in breast and prostate cancers. Blocking these molecular targets can initiate cell death or stop cell migration in the cancers.

A major component of their strategy is called structure-based, computer-aided design, a relatively new technology in the drug discovery field. Before ever working with an actual compound, the scientists can make manipulations to computer-designed molecules and observe simulated interactions between molecules and proteins to predict which structural changes will make the most sense to pursue.

“Most of the interaction between our compound and the overactive protein comes from what are called hot spots on the protein’s surface,” said Chenglong Li, assistant professor of medicinal chemistry and pharmacognosy at Ohio State and an expert in computational chemistry. “For each spot, we can design small chemical fragments and link them together to make a molecule. This is what computer-aided design and modeling can do.”

Some of the most effective compounds have been tested for their effectiveness against human cancer cell lines – as well as whether they might be toxic to healthy cells. So far, the molecule favored by the researchers has a nearly 100-fold difference in toxicity to cancer cells vs. healthy cells, meaning it takes 100 times more of the compound to kill a healthy cell than it does to kill a cancer cell.

“My basic premise,” Wetherbee said, “is that you take a capable microörganism like Klebsiella and you put it through the gruelling test of being exposed to a broad spectrum of antibiotics and it will eventually defeat your efforts, as this one did.” Although Tisch Hospital has not had another outbreak, the bacteria appeared soon after at several hospitals in Brooklyn and one in Queens. When I spoke to infectious-disease experts this spring, I was told that the resistant Klebsiella had also appeared at Mt. Sinai Medical Center, in Manhattan, and in hospitals in New Jersey, Pennsylvania, Cleveland, and St. Louis.

Unlike resistant forms of Klebsiella and other gram-negative bacteria, however, MRSA can be treated. “There are about a dozen new antibiotics coming on the market in the next couple of years,” Moellering noted. “But there are no good drugs coming along for these gram-negatives.” Klebsiella and similarly classified bacteria, including Acinetobacter, Enterobacter, and Pseudomonas, have an extra cellular envelope that MRSA lacks, and that hampers the entry of large molecules like antibiotic drugs. “The Klebsiella that caused particular trouble in New York are spreading out,” Moellering told me. “They have very high mortality rates. They are sort of the doomsday-scenario bugs.”

‘DIY’ kidney machine saves girl

A baby dying from kidney failure was saved when her doctor designed and built her a dialysis machine from scratch in his garage.

The job of the kidneys is to ‘clean’ the blood, and if they fail, a dialysis machine can do this job instead.

However, Dr Coulthard, together with senior children’s kidney nurse Jean Crosier, devised a smaller version, then built it away from the hospital. Millie was connected to the machine over a seven day period, allowing her own kidneys to recover.

Rebecca, from Middlesbrough, said: “It was a green metal box with a few paint marks on it with quite a few wires coming out of it into my daughter – it didn’t look like a normal NHS one.

The machine is still in use, helping babies in similar circumstances to Millie, but Dr Coulthard told the Newcastle Journal newspaper that an official version was needed. “This machine is only being used on the tiniest, earliest babies where there is nothing else that can be done.

“But if we had a machine that we could use much more freely, then we would be able to deal with many more babies and have a much greater chance of saving lives.”

The internship is for between 10 to 20 hours per week and can be done anywhere, as long as you have a computer, internet connection, and telephone. The dates of the internship are flexible accepted on a rolling basis.

Why ‘Licking Your Wounds’ Actually Works

scientists found that histatin, a small protein in saliva previously only believed to kill bacteria was responsible for the healing.

To come to this conclusion, the researchers used epithelial cells that line the inner cheek, and cultured in dishes until the surfaces were completely covered with cells. Then they made an artificial wound in the cell layer in each dish, by scratching a small piece of the cells away.

In one dish, cells were bathed in an isotonic fluid without any additions. In the other dish, cells were bathed in human saliva. After 16 hours the scientists noticed that the saliva treated “wound” was almost completely closed. In the dish with the untreated “wound,” a substantial part of the “wound” was still open. This proved that human saliva contains a factor which accelerates wound closure of oral cells.

Because saliva is a complex liquid with many components, the next step was to identify which component was responsible for wound healing. Using various techniques the researchers split the saliva into its individual components, tested each in their wound model, and finally determined that histatin was responsible.

Finding the Host Genes Viruses Require

Flu-infected fly cells reveal dependencies of the virus

The new study is important because it demonstrates a rapid-fire technique for identifying host factors such as proteins and carbohydrates that a virus commandeers to successfully infect a cell. By exposing the virus’s dependencies, the Wisconsin team has uncovered a target-rich environment for influenza drug developers.

By working in fly cells, the Wisconsin team was able to deploy a technique to rapidly and selectively silence thousands of genes to see which were used by the flu virus. Screening a library of some 13,000 genes, the group identified more than 100 whose suppression in fly cells hindered the virus’s ability to successfully take over the cell and make new viruses.

Magnetic Movie was shot in NASA’s Space Sciences Laboratories at UC Berkeley for Chanel 4 in association with the Arts Council of England.

In Magnetic Movie, Semiconductor have taken the magnificent scientific visualisations of the sun and solar winds conducted at the Space Sciences Laboratory and Semiconducted them. Ruth Jarman and Joe Gerhardt of Semiconductor were artists-in-residence at SSL. Combining their in-house lab culture experience with formidable artistic instincts in sound, animation and programming, they have created a magnetic magnum opus in nuce, a tour de force of a massive invisible force brought down to human scale, and a “very most beautiful thing.”

Magnetic Movie is the aquavit, something not precisely scientific but grants us an uncanny experience of geophysical and cosmological forces.

I post often on examples of scientific inquiry in action. I think it is an important way to see how science works while searching for answers. The process is not a simple one, but after a solution is found it can often be presented as obvious. But while trying to find answers it is quite difficult.

How Prozac sent the science of depression in the wrong direction

But the success of Prozac hasn’t simply transformed the treatment of depression: it has also transformed the science of depression. For decades, researchers struggled to identify the underlying cause of depression, and patients were forced to endure a series of ineffective treatments. But then came Prozac. Like many other antidepressants, Prozac increases the brain’s supply of serotonin, a neurotransmitter. The drug’s effectiveness inspired an elegant theory, known as the chemical hypothesis: Sadness is simply a lack of chemical happiness. The little blue pills cheer us up because they give the brain what it has been missing.

There’s only one problem with this theory of depression: it’s almost certainly wrong, or at the very least woefully incomplete. Experiments have since shown that lowering people’s serotonin levels does not make them depressed, nor does it does not make them depressed, nor does it worsen their symptoms if they are already depressed.

In this sense, Prozac is simply a bottled version of other activities that have a similar effect, such as physical exercise.

It is jarring to think of depression in terms of atrophied brain cells, rather than an altered emotional state. It is called “depression,” after all. Yet these scientists argue that the name conceals the fundamental nature of the illness, in which the building blocks of the brain – neurons – start to crumble. This leads, over time, to the shrinking of certain brain structures, like the hippocampus, which the brain needs to function normally.

Nebraska Firm Expands Recall of Beef Products Due To Possible E. coli O157:H7 Contamination, USDA

Nebraska Beef, Ltd., an Omaha, Neb., establishment is expanding its June 30 recall to include all beef manufacturing trimmings and other products intended for use in raw ground beef produced between May 16 and June 26, totaling approximately 5.3 million pounds

FSIS advises all consumers to safely prepare their raw meat products, and only consume ground beef or ground beef patties that have been cooked to a safe internal temperature of 160º F. The only way to be sure ground beef is cooked to a high enough temperature to kill harmful bacteria is to use a thermometer to measure the internal temperature.

Also as a result of the investigation, on June 25 FSIS announced a recall of ground beef products sold at Kroger retail establishments in Michigan and in Central and Northwestern Ohio.

Another example of the questionable state of food safety in the USA.
