Gram-negative Bacteria Defy Drug Solutions

Deadly bacteria defy drugs, alarming doctors by Mary Engel

Acinetobacter doesn’t garner as many headlines as methicillin-resistant Staphylococcus aureus, the dangerous superbug better known as MRSA. But a January report by the Infectious Diseases Society of America warned that drug-resistant strains of Acinetobacter baumannii and two other microbes — Pseudomonas aeruginosa and Klebsiella pneumoniae — could soon produce a toll to rival MRSA’s.

The three bugs belong to a large category of bacteria called “gram-negative” that are especially hard to fight because they are wrapped in a double membrane and harbor enzymes that chew up many antibiotics. As dangerous as MRSA is, some antibiotics can still treat it, and more are in development, experts say.

But the drugs once used to treat gram-negative bacteria are becoming ineffective, and finding effective new ones is especially challenging.

For the most part, gram-negative bacteria are hospital scourges — harmless to healthy people but ready to infect already-damaged tissue. The bacteria steal into the body via ventilator tubes, catheters, open wounds and burns, causing pneumonia, urinary tract infections, and bone, joint and bloodstream infections.

Pseudomonas is widely found in soil and water, and rarely causes problems except in hospitals.

I am not a fan of lifestyle drugs. People seem to forget that drugs have side effects that are quite large. Being surprised when a drug has adverse consequences shows a failure to understand the risks. You should assume adverse effects and take them only when that risk is outweighed by significant tangible benefits. A Boom in Memory-Enhancing Drugs?

A whole generation has come of age using attention-deficit drugs such as Adderall and Ritalin, a category valued at nearly $4.7 billion in 2007. A lot of teenagers have used them casually as study aids, often buying them on the Internet. And now, overworked professionals are seeing the appeal.

And when they almost certainly have significant adverse effects on many people, then people will get upset. Granted the drug companies pushing sales for negligible benefits do deserve condemnation. However, the larger problem is people that choose to risk their health as though they don’t have the ability to learn and can just ignore evidence of risks.

Study Finds Obesity as Teen as Deadly as Smoking

A Swedish study has found obesity is as deadly (increasing adult mortality) as smoking for teens. Combined effects of overweight and smoking in late adolescence on subsequent mortality: nationwide cohort study

Regardless of smoking status, overweight and obesity in late adolescence increases the risk of adult mortality. Obesity and overweight were as hazardous as heavy and light smoking, respectively, but there was no interaction between BMI and smoking status. The global obesity epidemic and smoking among adolescents remain important targets for intensified public health initiatives.

Obesity Just as Risky for Teens as Heavy Smoking

Dr. Martin Neovius of the Karolinska Institute and colleagues analyzed data from more than 45,000 18-year-olds who underwent military conscription tests in which their body mass index (BMI) and smoking status were recorded. The men were followed for an average of 38 years.

During the follow-up period, 2,897 of the men died. The incidence of death was highest among obese men and lowest among those of normal weight. Compared with those who had a normal weight at age 18, those who had been overweight were about a third more likely to die early, whereas those who were obese were more than twice as likely to die prematurely.

“To make a change in how you look, you are talking about a significant period of training,” Dr. Kraemer said. “In our studies it takes six months to a year.” And, he added, that is with regular strength-training workouts, using the appropriate weights and with a carefully designed individualized program. “That is what the reality is,” he said.

And genetic differences among individuals mean some people respond much better to exercise than others

Now, said Mr. Antane, who runs with a group in Princeton on Thursday nights, “everything changed — my outlook on life, who I hung out with, how I felt about myself.”

Our bodies evolved under conditions with much more exercise than we currently get if we sit in an office all day. And we had less food. It is no surprise with more food and less exercise that we gain weight. And given that the benefit of fat was to help us survive when we had little food out bodies don’t change overnight. If they did then our ancestors would have had much more difficulty surviving – the whole point was to provide a resource to tap in bad times. If that resource dissipated quickly it would not have helped much.

Moving Closer to Robots Swimming Through Bloodsteam

James Friend, of Monash University, said that such devices could enter previously unreachable brain areas, unblocking blood clots, cleaning vessels or sending back images to surgeons. “The first complete device we want to build would have a camera,” Professor Friend said.

Professor Friend said they had shown the motor, which is a quarter of a millimetre wide, had enough power to navigate this type of nanorobot through the bloodstream of a human artery. Tests of their prototype device in a liquid as viscous as blood were also promising. “It swam.”

The team plans to conduct animal tests of a nanorobot driven by their motor later this year or early next year. But Professor Friend cautioned that many technical hurdles needed to be overcome.

Their miniature motor was connected to an electricity supply and a way would need to be found to power it remotely. The construction of the flagella also needed refinement.

Correlation is Not Causation: “Fat is Catching” Theory Exposed

Their study was reported to have shown that you can “catch” obesity from having fat friends and that obesity is so contagious, it can be spread long-distance by email and instant messaging. Even healthcare professionals, who didn’t understand the etiology of true obesity or how statistics can be misused, failed to detect the implausibility of “second-hand obesity.” In fact, some doctors became so enamored with the new “science of networking” they believed it should be a new medical specialty: network medicine.

Jason M. Fletcher, Ph.D., assistant professor at the Yale School of Public Health in New Haven, Connecticut, along with Boston economist, Ethan Cohen-Cole, Ph.D., designed an ingenious study. They selected conditions that no one would seriously believe were spread by social networking and online friendships: height, headaches and acne. They then applied the same standard statistical methods used in Christakis and Fowler’s social networking research to “find” that acne, height and headaches have the same “social network effect.”

As they explained, patterns of association among people can lead to correlations in health conditions between friends that are not caused by direct social network effects at all.

There is a need for caution when attributing causality to correlations in health outcomes between friends using non-experimental data. Confounding is only one of many empirical challenges to estimating social network effects.

Excellent reminder of the risks of analyzing data for correlations. We continue to, far to often, fail to interpret data properly. Both authors of the study, received PhD’s from the University of Wisconsin-Madison which strengthens my belief that it is teaching students well (just kidding).

Also another example of the scientific inquiry process where scientists challenge the conclusions drawn by other scientists. It is a wonderful system, even if confusing and not the clean idea so many have of how science works.

Diabetes Up 90% in USA Since 1997

Diabetes Up 90% in U.S.

Type 2 diabetes is up 90% since 1997. And that may be an underestimate because the numbers come from self-reported surveys conducted by the CDC in 1995-1997 and in 2005-2007. About a third of people with diabetes don’t yet know they have the dangerous disease.

“The growth in diabetes prevalence has been concomitant with growth in obesity prevalence,”

Obesity, the CDC says, is the major risk factor for diabetes. Yet it’s not necessary to become thin to avoid this debilitating disease. A study of people at high risk for diabetes shows you can cut your risk of diabetes by 58% in a three-year period by doing just two things:

* Lose 5% to 10% of your body weight.
* Five days a week, get 30 minutes of moderate physical activity.

Lessons from the Amish: We’re not doomed to obesity

Four years ago we discovered that the Amish maintained super-low obesity levels despite eating a diet high in fat, calories and refined sugar. They key was their level of physical activity — men averaged 18,000 steps a day, women 14,000. That’s monumental compared to the paltry couple of thousand or so most of us eke out in a day.

A recent study revealed even more about the Old Order Amish: They maintain low obesity levels despite having a gene variation that makes them susceptible to obesity. The secret here? You guessed it — lots of physical activity.

Study Conclusions: “Our results strongly suggest that the increased risk of obesity owing to genetic susceptibility by FTO variants can be blunted through physical activity. These findings emphasize the important role of physical activity in public health efforts to combat obesity, particularly in genetically susceptible individuals.”

Sometimes the simple explanation is worth paying attention to. Add lack of activity to eating more (Obesity Epidemic Explained – Kind Of: 1970 Americans ate an average of 2170 calories per day in 2000 they ate an average of 2700) and it seems like it is logical we would gain weight due to these two factors.

Why ‘Licking Your Wounds’ Actually Works

scientists found that histatin, a small protein in saliva previously only believed to kill bacteria was responsible for the healing.

To come to this conclusion, the researchers used epithelial cells that line the inner cheek, and cultured in dishes until the surfaces were completely covered with cells. Then they made an artificial wound in the cell layer in each dish, by scratching a small piece of the cells away.

In one dish, cells were bathed in an isotonic fluid without any additions. In the other dish, cells were bathed in human saliva. After 16 hours the scientists noticed that the saliva treated “wound” was almost completely closed. In the dish with the untreated “wound,” a substantial part of the “wound” was still open. This proved that human saliva contains a factor which accelerates wound closure of oral cells.

Because saliva is a complex liquid with many components, the next step was to identify which component was responsible for wound healing. Using various techniques the researchers split the saliva into its individual components, tested each in their wound model, and finally determined that histatin was responsible.

I actually thought that they were a lot about science. That’s what they tell the public. They are all about science and discovering new drugs. But as I started to follow their daily activities and talk to executives, I learned that really it was marketing that drove them.

According to Petersen, the rewards have been large. America has become the top consumer of prescription drugs in the world, with nearly 65% of the population on physician-prescribed medication. In 2005, Americans spent $250 billion dollars on such drugs. This consumption made pharmaceuticals the most profitable business sector in America from 1995-2002.

We’ve come to a time when decisions on how to treat a disease have as great a chance of being hatched in a corporate marketing department as by a group of independent doctors working to improve the public’s health.

Unfortunately patients are driven more by marketing than medicine. Much worse though, doctors seem to bend to these patients marketing driven desires. Plus the corrupting influence of money on research and marketing to doctors seems likely a significant reason for the poor performance and high cost of USA health care.
